Potential new therapeutic options include anticancer peptides and proteins derived from antimicrobial peptides. The purpose of this study was to evaluate the cytotoxic effect and the cell death mechanism of the hypothetical protein PHP-1 of the psychrophilic microorganism Polaromonas sp. in the cervical cancer cell line. Through bioinformatic analysis, it was shown that PHP-1 exhibits structural characteristics typical of anti-cancer peptides and proteins. The cytotoxicity of PHP-1 was evaluated by an MTT test that showed a dose and time-dependent effect. The effects generated were additionally analyzed by means of an annexin V / PI labeling, indicating that the hypothetical protein induces apoptosis, which correlates with the observed morphological changes. The results of this work represent the first functional study of the hypothetical protein PHP-1 and suggest that PHP-1 is cytotoxic for the HeLa cell line by inducing cell apoptosis, highlighting its potential as an anticancer agent. |

El cáncer del cuello uterino presenta un alto índice de mortalidad entre las mujeres, siendo una de las neoplasias malignas con mayor incidencia. Las opciones terapéuticas actuales son altamente invasivas; por esto son necesarias alternativas de tratamiento con menores efectos secundarios e invasividad. Entre las nuevas posibles opciones terapéuticas se encuentran los péptidos y proteínas anticancerígenas derivadas de péptidos antimicrobianos. El propósito de este estudio fue evaluar el efecto citotóxico y el mecanismo de muerte celular de la proteína hipotética PHP-1 del microorganismo psicrófilo Polaromonas sp. en la línea celular de cáncer cérvico-uterino. Por medio de análisis bioinformáticos, se demostró que la PHP-1 exhibe características estructurales propias de los péptidos y proteínas anticancerígenas. La citotoxicidad de la PHP 1 fue evaluada mediante un ensayo MTT que mostró un efecto dosis y tiempo dependiente. Los efectos generados fueron complementariamente analizados por medio de una marcación de anexina V/PI, indicando que la proteína hipotética induce apoptosis, lo cual correlaciona con los cambios morfológicos observados. Los resultados de este trabajo representan el primer estudio funcional de la proteína hipotética PHP-1 y sugieren que la PHP-1 es citotóxica para la línea celular HeLa mediante la inducción de apoptosis celular, destacando su potencial como agente anticancerígeno.
